It's one of the most eagerly anticipated dates of the football calender by fans. For the big clubs the road to Wembley begins, while for lower league clubs they hope that their Cup run can go on just a little bit further by knocking out a giant.
Four non league clubs remain in the FA Cup, with the most high profile tie of the round taking place at Field Mill as Conference Premier outfit Mansfield Town take on seven time winners and last seasons runners up Liverpool.
Stags manager Paul Cox said: "I think the whole town is up for it. We want to make a good account of ourselves and make the supporters proud of our players. It's a fabulous time at the football club."
Elsewhere, Luton Town welcome Championship side Wolves to Kenilworth Road, with the visitors out of form having won only one of their last five games and manager Stale Solbakken said to be under pressure.
Another Conference Premier side host Championship opposition as Macclesfield Town play Cardiff City at Moss Rose. The Silkmen dropped from the football league last season but welcome the Championship leaders looking to upset the form guide.
The lowest team left in the Cup are Hastings United. The Sussex based club ply their trade in the Isthmian League Premier Division, the seventh tier of the English football pyramid, and face the long trip to Teeside to face Championship side Middlesbrough.
There are four all Premier League ties, including holders Chelsea who travel to Southampton looking to bounce back from their surprise 1-0 home defeat against Queens Park Rangers on Wednesday.
The R's will be in confident mood following that win, but there will be no respite as they host West Bromwich Albion at Loftus Road.
Premier League leaders Manchester United had a fantastic Christmas, winning three and drawing one of their four games over the festive period and extending their lead at the top to seven points. If they are to lift the FA Cup for the first time since 2004 in May, they will have to overcome a tricky tie at Sam Allardyce's West Ham.
The final all top flight tie takes place in Wales, as Swansea host Arsenal. The Swans will be hoping for a repeat of their 2-0 win over the Gunners earlier this season at the Emirates, while the FA Cup is the last realistic chance for Arsenal to win their first trophy since 2005.
There are several Premier League teams looking to avoid potential banana skins against Championship clubs, including out of form Newcastle United who travel to Brighton. These two sides met in the fourth round last season, with the Championship side winning 1-0.
Elsewhere, Sunderland travel to Bolton Wanderers, Norwich make the short trip to Peterborough United and Stoke go to Crystal Palace.
Another out of form Premier League side Aston Villa face Ipswich at Villa Park while 2011 winners Man City host Watford.
